Dodecyl maltoside (DDM) is a non-ionic detergent that consists of a hydrophilic maltose head and a hydrophobic long chain alkyl tail. It has a relatively low critical micelle concentration of 0.17 mM and is considered a gentle but powerful detergent. DDM is often the best tool for solubilising/crystallising membrane proteins. Membrane proteins usually have α-helical structures that are easily destroyed when the protein is released from its membrane environment. DDM can often preserve these structures during the solubilisation. In addition, membrane proteins can often be renatured when isolated with DDM.
n-Dodecyl-β-D-maltoside (DDM) is a non-ionic detergent commonly used to solubilize membrane-associated proteins. In particular, it helps retain the native conformation and activity of membrane-associated proteins, and facilitates the reforming of these proteins after denaturation. The critical micelle concentration of DDM is approximately 0.18 mM in water, decreases in the presence of sodium chloride or sucrose, and increases in urea.
DDM is the most widely used detergent for the isolation and purification of GPCR’s.
Dodecyl beta-D-maltoside is a glycoside resulting from attachment of a dodecyl group to the reducing-end anomeric centre of a beta-maltose molecule. It has a role as a detergent. It is a glycoside and a disaccharide derivative. It derives from a hydride of a beta-maltose.
